The senescence-accelerated prone mouse (SAMP8): A model of

age-related cognitive decline with relevance to alterations of the gene

expression and protein abnormalities in Alzheimer’s disease

SAM was estab in 1981... characteristic learning and memory deficitis at old age.

Selective breeding was applied based on the degree of senescence, the lifespan, and

age-assoc pathologic phenotypes.

SAMP8 mice have charac. learning and memory deficitis at old age.

Look at Table 1: they are 10 difft SAM strains... all with difft deficits:

amyloidosis, impaired immunity

degen arthirts

senile osteoporosis

age-depend amyloid

age-related cataracts

brain atrophy, contracted kidney

cancers, colitis.

incr oxidative stress

finding consistent with free radical theory of aging that posits the oxidative modification by reactive oxidative species (ROS).

Alterations in the permeability of the BBB blood brain barrier could contrib. to decline in energy metab in age SAMP8 brain.

This review indicates that oxidative stress is critic. important in these processes and amyloid beta may play a signif role

in agre-rel cognit. decline.
