Lead, & Hormones
Lead ingestion can affect various hormones in both humans and animals. Lead is considered an endocrine-disrupting chemical (EDC) due to its interference with hormone biosynthesis, metabolism, or action.
Effects on Male Reproductive Hormones
Lead exposure has been shown to disrupt the male reproductive endocrine system.
Gonadotropin-Releasing Hormone (GnRH) and Luteinizing Hormone (LH)
- Studies in male rats have demonstrated that both short-term high-level and long-term low-dose lead exposure can alter hypothalamic GnRH at the molecular level.
- Long-term, low-dose lead exposure in rats significantly increased GnRH mRNA concentrations, although the increase was attenuated at higher lead concentrations with increased exposure duration.
- Occupational exposure to lead in human workers resulted in significantly higher plasma LH and follicle-stimulating hormone (FSH) levels, particularly in workers exposed to moderate levels of lead (mean current blood lead concentration of 35.2 µg/dL).
- However, one rat study found no significant differences in serum LH among groups treated with various lead acetate concentrations for different time periods.
Testosterone
- In male rats, lead exposure has been linked to suppressed testosterone levels.
- A study on Wistar rats showed a significant decrease in testosterone levels after lead acetate ingestion.
- The effect of lead on testosterone levels was more pronounced in older animals, indicating an age-related impact.
- While occupational lead exposure in human workers did not show a significant overall difference in testosterone, older exposed workers (40 years and above) had significantly lower plasma testosterone concentrations.
- Workers exposed for 10 years or more also exhibited significantly lower testosterone levels compared to non-exposed workers.
Effects on Female Reproductive Hormones
Lead ingestion can also impact female reproductive hormones.
Follicle-Stimulating Hormone (FSH), Luteinizing Hormone (LH), Estradiol, Progesterone, and Testosterone
- A study on female Wistar rats exposed to lead acetate in drinking water for 12 months showed several changes in hormone levels.
- FSH levels were significantly decreased, though still within physiological limits.
- Serum LH levels were significantly higher.
- Estradiol and progesterone levels were significantly decreased.
- Serum testosterone levels were significantly higher in direct correlation with exposure levels.
- Another study in adult female BALB/c mice found that lead acetate caused a decrease in serum estrogen and progesterone levels.
Effects on Thyroid Hormones
The impact of lead on thyroid hormones is less consistent across studies.
- Some animal studies, such as one on male mice, indicated that lead exposure could alter T3 and T4 hormone levels.
- However, a meta-analysis of studies investigating occupational lead exposure on thyroid hormones in humans found no significant differences in mean thyroid hormone concentrations (including thyroid-stimulating hormone, total and free thyroxine, and total and free triiodothyronine) between exposed and control groups. This meta-analysis also found no relationship between blood lead or duration of exposure and thyroid hormone concentrations in men.
Effects on Calcitropic Hormones
Lead exposure can affect hormones involved in calcium regulation.
- Occupational lead exposure has been associated with significantly higher levels of parathyroid hormone (PTH) and 1,25-dihydroxyvitamin D (calcitriol).
- These increases are considered a compensatory mechanism to maintain serum calcium levels within a normal range.
- Lead indirectly alters bone cell function through changes in circulating levels of hormones like 1,25-dihydroxyvitamin D3.
General Endocrine Disruption
Lead is recognized as an endocrine-disrupting chemical that can interfere with the normal balance of endocrine systems. Its mechanisms involve various pathways, including estrogenic, antiandrogenic, thyroid, and other nuclear receptor actions, as well as effects on steroidogenic enzymes and neurotransmitter systems. The multisystem toxic effects of lead suggest it satisfies the conditions defining EDCs.
