How to show liver essentiality
What would be needed to make the liver-stage claim
Pharmacology first (cheapest). NH125 (or a more selective analog) against P. falciparum or P. berghei liver stages in primary human hepatocytes / HC-04 / HepG2, with a sporozoite-invasion arm to separate invasion from intrahepatic development. A causal, if indirect, readout that needs no new parasite line.
Conditional KO carried through transmission. The existing DiCre line is rapamycin-inducible, so an unexcised population is transmission-competent; induce after sporozoite production (in sporozoites or in infected hepatocytes) and score liver-stage development in FRG huHep mice or hepatocyte culture. Note the caveat that PfSnf2L depletion reduces sexual conversion, so gametocyte and sporozoite yields from the parental line need checking, and excision efficiency must be verified in sporozoites/EEFs rather than assumed.
Rodent-model equivalent. A conditional PBANKA_0942700 line (DiCre, knock-sideways, or promoter swap) transmitted to salivary-gland sporozoites, with depletion induced at sporozoite/liver stage; a straight KO is excluded by the blood-stage essentiality above.
Protein-level confirmation in sporozoites/EEFs (IFA or tagged line) — the evidence in section 2 is transcript-level only; sporozoite proteome datasets were not examined here.
How to phrase it in the meantime
Defensible: "PfSnf2L is essential for asexual blood-stage development and early gametocytogenesis (PMID 39972139) and is transcribed at high level in sporozoites and throughout liver-stage development (PMID 39891010, GSE220039); its requirement in pre-erythrocytic stages has not yet been tested."
