Two-photon excitation microscopy

Apr 28, 2011 09:32 PM pioneered by Winifred Denk patented by him and lic. to Carl Zeiss.

allows micro of living tissue with fluor up to depth of about 1 mm.

two photons of comparably lower energy than needed for one photon excitation can also excit

a fluorophore in one quantum event.

because it requires confluence of two laser beams, so excitation is restricted to a tiny focal volume

like 1 femtoliter. so excitation is highly localized.
